Plasmid production is fundamental to AAV manufacturing, using plasmids as the initial raw material for subsequent mammalian cel transfection. Our in-house plasmid production platform facilitates this process. Upstream production of distinct plasmids occurs in 20L or 100L bioreactors, yielding kilogram-scale wet cell mass. Before the plasmids are purified chromatographically, the cells need to be broken up. This is done by alkaline cell lysis. Current research aims to optimize this step by implementing continuous alkaline cell lysis using a plug flow reactor, characterized by a defined residence time distribution and low shear environment. Additionally, dynamic mixers are employed to homogenize process streams, preventing extreme pH exposure when using concentrated buffers. This approach minimizes the handling of large buffer volumes and therefore intensifies the following downstream process.
